Branco, Marcelo, Miriam Ribeiro, Nubio Negrão, and Antonio C. Bianco. 3,5,38-Triiodothyronine actively stimulates UCP in brown fat under minimal sympathetic activity. Am. J. Physiol. 276 (Endocrinol. Metab. 39): E179–E187, 1999.—To investigate the role of type II 58-deiodinase (58D-II) in the expression of mitochondrial uncoupling protein (UCP) in brown adipose tissue (BAT), we injected intact male rats with reverse (r) 3,5,38-triiodothyronine (T3; 100 μg·100 g body wt21 ·day21), an inhibitor of 58D-II, for 2–5 days. UCP decreased by ,20% in rats kept at 28°C and failed to increase during cold exposure (4°C). Next, thyroxine treatment (1–10 μg·100 g body wt21 ·day21) increased nuclear T3 in rats kept at 28 or 4°C. In these rats, nuclear T3 correlated positively with UCP. In addition, T3 (1–50 μg·100 g body wt21 ·day21) given to intact rats (5–15 days; 28°C) induced an approximately twofold increase in UCP. In these T3-treated animals, the interscapular BAT thermal response to norepinephrine infusion also correlated positively with T3 dose and UCP content. Treatment with propranolol or reserpine failed to block the T3 induction of UCP (,1.8and ,2.3-fold). The results emphasize the importance of local 58D-II and reveal an independent role of T3 in the expression of UCP.

To investigate the role of type II 5'-deiodinase (5'D-II) in the expression of mitochondrial uncoupling protein (UCP) in brown adipose tissue (BAT), we injected intact male rats with reverse (r) 3,5,3'-triiodothyronine (T3; 100 μg ⋅ 100 g body wt-1 ⋅ day-1), an inhibitor of 5'D-II, for 2-5 days. UCP decreased by ∼20% in rats kept at 28°C and failed to increase during cold exposure (4°C). Next, ...

Triiodothyronine amplifies the adrenergic stimulation of uncoupling protein expression in rat brown adipocytes

Hernández, Arturo, and Maria Jesús Obregón. Triiodothyronine amplifies the adrenergic stimulation of uncoupling protein expression in rat brown adipocytes. Am J Physiol Endocrinol Metab 278: E769–E777, 2000.—Uncoupling protein (UCP), the mitochondrial protein specific to brown adipose tissue, is activated transcriptionally in response to cold and adrenergic agents. We studied the role of triiod...

Growth factor regulation of uncoupling protein-1 mRNA expression in brown adipocytes.

To study the effect of the mitogens epidermal growth factor (EGF), acidic and basic fibroblast growth factors (aFGF and bFGF), and vasopressin on brown adipocyte differentiation, we analyzed the expression of uncoupling protein-1 (UCP-1) mRNA. Quiescent brown preadipocytes express high levels of UCP-1 mRNA in response to triiodothyronine (T3) and norepinephrine (NE). The addition of serum or th...

Triiodothyronine amplifies the adrenergic stimulation of uncoupling protein expression in rat brown adipocytes.

Uncoupling protein (UCP), the mitochondrial protein specific to brown adipose tissue, is activated transcriptionally in response to cold and adrenergic agents. We studied the role of triiodothyronine (T(3)) on the adrenergic stimulation of UCP mRNA expression by use of primary cultures of rat brown adipocytes. Basal UCP mRNA levels are undetectable. Norepinephrine (NE) increases UCP mRNA during...

Differences in the response of UCP1 mRNA to hormonal stimulation between rat and mouse primary cultures of brown adipocytes.

UNLABELLED Uncoupling protein 1 (UCP-1), the specific marker of brown adipose tissue, is transcriptionally activated in response to adrenergic stimuli and thyroid hormones are necessary for its full expression. We describe differences in the regulation of UCP-1 mRNA expression between rat and mouse brown adipocytes in culture, using norepinephrine (NE), triiodothyronine (T3), insulin and retino...
